Lipidomics Core Facility

Mission

The Lipidomics Core Facility is a research facility located at the Department of Biochemistry and Molecular Biology, at the University of Debrecen, Faculty of Medicine. The laboratory’s mission is to develope and optimize SRM-based targeted lipid identification and quantification methods with high emphasis on lipid metabolites enabling research principally in the Faculties of Science and Medicine of the University of Debrecen. The Lipidomics Core Facility undertakes research, collaborations and analytical services for other Universities and biotech companies in the lipidomics field providing access to state-of the art lipid metabolite identification and quantification.

Instrumentation

Waters 2695 HPLC coupled to Micromass Quattro Ultima Platinium (Waters) triple quadrupole-type mass spectrometer and Waters 2998 Photodiode Array Detector operated by MassLynx V4.1 (Waters) software

Skyline software for evaluation of SRM data

Services

Eicosanoid analysis

The following molecules can be examined parallely: arachidonic acid, prostaglandins such as PGE2, PGD2, PGF, 6-keto PGF, PGA2, PGB2, PGJ2 and thromboxane B2 (TXB2), 5-HETE and 15-HETE, oxidized lipids such as 9-HODE and 13-HODE

  1. Analysis of pro- and anticoagultion balance: determination of PGI2:TXA2 ratio by analysing the amount of their stable metabolites, the 6-keto PGForiginating from the instable PGI2 and the TXB2 originating from instable TXA2
  2. Analysis of inflammation: quantification of different prostaglandins
  3. Oxidized lipid quantification: quantification of 9-HODE and 13-HODE

Endocannabinoid analysis

Quantitative analysis of anandamide, 2-arahcydonyl glycerol (2-AG), 1-arahcydonyl glycerol (1-AG), oleoyl-etanolamide (OEA) and palmytoyl-ethanolamide (PEA)

Method development for lipid metabolite analysis

Sample types for analysis

Serum, plasma, cell culture supernatants, cell lysates, animal or human tissues/tissue lysates, biopsies

Requirements

100 mg tissue or 100 µl liquid and minimum 5 samples to be analysed. For endocannabinoid analyses minimum 5 million cells are required. These amounts are for reference only, depending on the type of analyses and type of sample they can vary.

Sample submission

Samples should be submitted along with a filled and signd sample request form to University of Debrecen, Theoretical building, Room 205. Before sample submission, please contact Dr. Eva Csősz.

Conditions of Use

After sample analysis the BMBI Lipidomics Core Facility sends to the Costumer a report and a bill containing the costs of the analyses. The report will be sent in electronic and printed formats as well. The printed format needs to be signed by the Costumer and should be sent back to the BMBI Lipidomics Core Facility for record keeping purposes. You should note that the payment for the analyses is due even if the results generated are not the expected ones, but control runs prove that the equipment worked well and the analysis was done properly!

An appropriate acknowledgement of the involvement of the BMBI Lipidomics Core Facility shall appear in any publication involving work in the BMBI Lipidomics Core Facility. The following wording is suggested:

"Mass spectrometry analyses for this work were carried out at the BMBI Lipidomics Core Facility, Department of Biochemistry and Molecular Biology University of Debrecen."

Note also that often other grants/support would also need to be acknowledged.

If the BMBI Lipidomics Core Facility is involved in assay development and/or validation work for the project, co-authorship is required with the appropriate member(s) of the lab.

A copy (or full citation details) of any publication involving work in the BMBI Lipidomics Core Facility should be sent to the facility manager as soon as possible after publication.

Submission of an analysis request form provides your agreement to the above conditions.

Costs

Costs are discounted for groups from University of Debrecen, Faculty of Medicine (and its affiliated institutes). Commercial fees are competitive and compare favourably with costs for similar services from other laboratories.
